Gene Expression Profile in Fetal Bovine Serum-Induced Abnormal Differentiation of Skin Keratinocytes

Abstract
The proliferation and abnormal differentiation of dermal keratinocytes in psoriasis are reported. The exposure of keratinocytes to 5% serum in keratinocyte growth medium(KGM) leads to differentiation with the expression of SKALP, a biomarker for psoriasis differentiation. This study established a possible mechanism of abnormal differentiation based on the gene expression profiles of keratinocytes in the presence of 5% serum, one differentiation inducer. Keratinocytes were stimulated by a single exposure to KGM containing 5% fetal bovine serum(FBS); simultaneously, other cells were incubated with KGM in the absence of growth factors. After comparing the gene expression profiles using a microarray, the cellular events during differentiation were predicted from the changes in gene expression. The expression of the differentiation markers keratins 1 and 10 was significantly suppressed in the presence of 5% FBS, and this was accompanied by morphological changes over time. The microarray analysis revealed that the transcription of inflammatory cytokines and signal transduction molecules increased rapidly, while genes encoding precursor proteins and proteolysis were increased in the late stage of differentiation. During abnormal differentiation, keratinocytes express various inflammatory cytokines, growth factors, and chemokines in the early stage. The final differentiation takes place more rapidly than the normal differentiation process with the expression of SKALP, a psoriatic differentiation marker.
